WHAT ARE THE BENEFITS OF VAPOR PHASE (-150°C TO -190°C) LIQUID NITROGEN STORAGE?
Storing cryovials in vapor-phase liquid nitrogen provides increased safety, while also providing increased accessibility to samples.
Vapor phase storage eliminates the very rare occurrence of cross-contamination from viral samples submerged in liquid nitrogen. Although Lisa Cryobanking and our affiliates have never experienced this problem, we store viral and bacteria stocks in a separate dewar.
Cell activity effectively stops at -135°C. As all biological activity is ceased, the cells are left in a state of suspended animation - preserving them perfectly from the moment they were frozen.
WHAT SAMPLES ARE BEST FOR VAPOR PHASE STORAGE?
Cell lines, plasma, viral stocks, bacterial stocks, yeast stocks, mouse tissue, RNA... Just about everything!
Any samples or cryoboxes subject to retrieval and opening in the next 12 months should also be kept in vapor phase.
Properly controlled vapor phase storage can be just as effective as liquid phase.

HOW ARE CELLS FROZEN DOWN SUCSESSFULLY?
Ideally, healthy proliferating cells during a passage are frozen down in full media ( ex. RPMI + 10% FBS), in a cryoprotectant such as DMSO.
Cells should be frozen down at -1°C/min in a -80°C freezer before being shipped to Lisa Cryobanking on dry ice.
Live cells and yeast can be sent to Lisa Cryobanking in the appropriate culture dish at 60-70% confluence. We can then test, propagate and freeze down your active cultures.
